Özet

Transcription of the yeast retrotransposon Ty1 is regulated by a large set of transcription factors and chromatin modifying complexes ina cell type dependent manner. In this study, we have shown that the transcription of Ty1 is activated by low glucose signaling in diploid S.cerevisiae cells. Growth of diploid yeast cells in low glucose medium lead to at least 3-fold activation of Ty1 transcription. Our resultsindicated that this activation largely depends on the transcription factor Tup1p. In addition to Tup1p, Snf2p, one of the subunits of chromatinmodifying complex Snf/Swi, is also essential for the regulation of Ty1 transcription. Moreover, our results indicated that the transcription ofTy1 in haploid S. cerevisiae cells were not affected by glucose signaling and remained at same levels in high glucose or in low glucosemedium. In tup1 mutant diploid yeast cells, transcription of Ty1 remained at the basal level and it is not regulated by glucose signaling.These results indicated that transcription of Ty1 is regulated differentially by glucose signaling in a cell type, Tup1p and Snf2p dependentmanner
