Formation of Biocompatible Star Polymeric Nanoparticles with a Double Decker Phthalocyanine Core

Phthalocyanines have attracted significant attention due to their unique properties and potential diverse applications, particularly in biomedical applications such as photodynamic therapy and chemical biosensing. However, most phthalocyanines are hydrophobic and tend to aggregate in aqueous solutions, which limit their efficacy in biological systems. One possible solution to overcome this drawback is to encapsulate phthalocyanines in biocompatible polymeric nanoparticles.

In this work, double-decker phthalocyanines with an “octaSR” substitution pattern and 16 hydroxyl functions were synthesized and used as a precursor for the ROP (ring-opening polymerization) of ε-caprolactone with different poly(ε-caprolactone) (PCL) chain lengths, as previously reported with a near-infrared absorbing photosensitizing phthalocyanine [1]. The resulting star polymers with 16 PCL arms and with the double-decker phthalocyanine core have been converted into nanoparticles by the nanoprecipitation technique, yielding biocompatible nanoparticles suitable for biomedical applications. The different conditions used to prepare the nanoparticles, as well as their structural and spectroscopic characterization will be presented.
