Özet

Amaç: Oreksin, oreksin-1 ve oreksin-2 reseptörlerini etkileyen; yalnızca lateral hipotalamustaki nöronlardan üretilen uyku/uyanıklığı düzenleyen bir nöropeptidtir (1, 2). Oreksin reseptörleri, kemirgen beynindeki katman 6b kortikal nöronlarda güçlü bir şekilde eksprese edilir ve bu nöronların bazıları geniş intrakortikal ve talamik projeksiyonlara sahiptir. Uyku regülasyonu ile ilgili çalışmalarda l aboratuvar ortamında geliştirilen YNT-185, non-peptid oreksin tip-2 reseptör agonistidir. Çalışmamızda; YNT-185’in intraserebroventriküler (ISV) enjeksiyonu ile Strasbourg orijinli genetik absans epilepsili sıçanlarda (GAERS) görülen diken-ve-dalga deşarjlarının (DDD) üzerinde düzenleyici bir rolü olabileceği hipotezini test ettik.

YöntemlerBu çalışmada, ketamin (100 mg/kg, i.p) ve ksilazin (10 mg/kg, i.p) anestezisi altında stereotaksik cerrahi işlem uygulanarak ISV rehber kanül ile (AP:-1,0; ML:1,4; V:4,1 mm) bilateral elektroensefalografi (EEG) kayıt elektrodları GAERS (n=5) kafatasına yerleştirildi. Bir haftalık derlenme süresi sonrası hayvanlardan 3 saatlik bazal EEG kaydı alındı. Sonraki gün 100 nmol/10uL ISV YNT-185 enjeksiyonundan sonra 3 saat EEG kaydı alındı. EEG sinyalleri Powerlab 8S EEG kayıt sistemi ile kaydedildi ve LabChart 8.0 Windows programı ile analiz edildi. Paired t testi kullanıldı (p<0,05 anlamlı olarak kabul edildi).

Bulgular: YNT-185’in ISV enjeksiyonu sonrası DDD’lerin kümülatif süresinde ve her bir DDD’nin süresinde bazal kayda göre istatistiksel olarak anlamlı derecede azalma görülmüştür (p<0, 05).

Tartışma ve Sonuçlar: Bulgular, YNT-185’in GAERS’lerde görülen DDD oluşumunda bazal EEG kaydına göre azalmaya yol açtığını göstermektedir. GAERS'de orexinin ilk kez DDD'ler üzerindeki baskılayıcı etkisinin ve nöropeptiderjik absans nöbetleri üzerinde modüle edici bir etkiye sahip olabileceğini düşündürdüğünü gösterilmiştir. Buradaki zorluk, bu etkinin kortikal veya subkortikal düzeyde aracılık edip etmediğini belirlemektir. YNT-185’in DDD oluşumu üzerine etkisini daha iyi anlayabilmek için çalışmamızı kapsamlandırmayı hedeflemekteyiz.

Objectives: Orexin, a neuropeptide, regulates sleep and wake rhythms through orexin-1 and orexin-2 receptors and is produced from the lateral hypothalamus neurons (1, 2). Orexin receptors are strongly expressed in layer 6b cortical neurons in the rodent brain, and some of these neurons have extensive intracortical and thalamic projections. YNT-185 as a non-peptide orexin type-2 receptor agonist has been investigated in several studies related to sleep. In this study, we tested the hypothesis that YNT-185 might have a modulatory role on spike- and-wave discharges (SWDs) of genetic absence epilepsy rats from Strasbourg (GAERS).

Methods: We inserted stereotaxically an ICV guide cannula (AP:-1.0; ML: 1.4; V:4.1 mm) and bilateral electroencephalography (EEG) recording electrodes on skull of GAERS (n=5) under ketamine (100 mg/kg, i.p.) and xylazine (10 mg/kg, i.p.) anesthesia. After a one-week recovery period, 3-hour basal EEG recordings were obtained from animals. Next day, the dose of 100 nmol/10uL YNT-185 was administered. Before and after the injection, EEG signals were recorded with Powerlab 8S EEG recording system and analyzed with LabChart 8.0 Windows program. Paired t test was used (p<0.05 was considered significant).

Results: The injection of ICV YNT-185 statistically decreased the cumulative duration of SWDs and duration of each SWD when compared to baseline recording (p< 0.05).

Conclusions: The findings showed that YNT-185, a non-peptide orexin type-2 receptor agonist, reduces SWD compared to basal SWD in GAERS. The challenge is to determine whether this effect is mediated on cortical or subcortical level. We further continue this study in order to understand the effect of YNT-185 on SWD.
