Encapsulation of Photosensitising Phthalocyanines into Poly(Malic Acid) Based Nanoparticles: Structural and Experimental Effects

Phthalocyanines (Pcs) are among the most effective photosensi-tisers for photodynamic therapy, mainly due to their strong ab-sorption in the phototherapeutic window [1-2]. Most of them are hydrophobic, resulting in self-aggregation in aqueous media, which lowers their photodynamic efficiency. To overcome this obstacle, the encapsulation of Pcs into polymeric nanoparticles (NPs) to enhance their photophysico-chemical properties and solubility in water, as well as disrupt their aggregation is a good strategy [3]. Poly(malic acid) (PMLA) and its derivatives are good polymeric nanomaterials prepared via anionic ring-opening polymerization of benzyl malolactonate (MLABe). Two derivatives of PMLA, poly(benzyl malate) (PMLABe73) as a lipophilic polymer, and poly(ethylene glycol)-b-poly(benzyl malate) (PEG42-b-PMLABe73) as an amphiphilic block copolymer have been selected. To achieve tumor-targeting, biotin-functionalized Biot-PMLABe73 and Biot-PEG42-b-PMLABe73 were also prepared [4-5]. Three hydrophobic Pcs metalated with Zn or Pd and with either linear or bulky substituents (PdPc-OHex, ZnPc-OHex, and PdPc-OEtHex) were encapsulated into the hy-drophobic polymeric core with systematic variations of the encapsulation conditions.
