Final workshop of VascAgeNet, Riga, Letonya, 20 - 21 Mart 2024
Pharmacological modulation of endothelial dysfunction and arterial
stiffness as hallmarks of vascular ageing, holds
great promise for improving cardiovascular health in the ageing population.
The multi-faceted nature of vascular ageing
complicates the evaluation of drugs. Interactions with exercise training
and sex/gender-related differences, which should be taken into account
Pharmacological modulation of mechanisms and signalling pathways that
contribute to vascular ageing are involving
·
Oxidative stress
·
Renin-angiotensin-aldosterone system
· Cellular senescence
· Macroautophagy
·
Extracellular matrix remodelling
·
Calcification
·
Gasotransmitters
There are also approved drugs with off-label effects on vascular ageing and
we should not e that dietary supplements have some limited effects on vascular
ageing
At present, there are no approved drugs
specifically targeting arterial stiffness and endothelial dysfunction. Some available
cardiovascular and anti-diabetic drugs can modulate vascular ageing. Novel promising
drugs are currently evaluated in preclinical and clinical trials.
Further
research is needed to fully elucidate the underlying mechanisms and optimize
the efficacy and safety of these interventions for clinical translation.
